Layton Brothers: Mystery Room é um spin-off da série Professor Layton para os sistemas operacionais móveis iOS e Android. Foi anunciado no evento LEVEL-5 Vision de 2011. O jogo segue Lucy Baker, uma detetive iniciante transferida para o Scotland Yard. Aqui, ela trabalha sob Alfendi Layton, filho do renomado professor Hershel Layton. Juntos, eles investigam crimes para a "Mystery Room" do Scotland Yard. O jogo foi lançado como um freemium: o próprio jogo e os primeiros 2 casos (e prólogo) são gratuitos, enquanto dois pacotes adicionais de respectivamente 4 e 3 casos podem ser comprados dentro da aplicação.
