Last Specter é o quarto jogo da série Professor Layton e é uma prequela que se passa três anos antes do primeiro trilogia, detalhando como o professor Layton conheceu seu aprendiz, Luke Triton. O jogo inclui mais de 170 puzzles. Professor Layton and the Last Specter também inclui um additional role-playing game chamado Professor Layton's London Life, disponível desde o início do jogo. London Life, em que os jogadores interagem com vários personagens da série em uma cidade chamada "Little London", foi anunciado para conter mais de 100 horas de conteúdo. London Life foi removido das versões europeias do jogo a fim de evitar um atraso significativo na liberação do jogo devido à tradução.
