O 'Modo Aventura' de Fullmetal Alchemist: Brotherhood permite que os jogadores vivam a aventura dos irmãos Elric enquanto lutam para desvendar uma trama maligna e enfrentarem outros alquimistas poderosos em batalhas até à morte. O jogo apresenta um grande número de personagens adicionais personalizáveis, onde os pontos ganhos na batalha são usados para melhorar seus poderes especiais e habilidades. As lutas ocorrem em diferentes arenas de combate onde os jogadores podem exibir suas impressionantes capacidades alquímicas, usar a topografia do terreno estrategicamente para ganhar vantagem e lançar cuidadosamente ataques combinados especiais com seu parceiro de batalha para mudar o curso da luta.
